Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Each receptor contributes a different physiological effect. Activation of the GLP-1 receptor slows gastric emptying and reduces appetite signaling in the brain. GIP receptor activity influences insulin secretion and lipid handling, while glucagon receptor stimulation raises energy use and fat oxidation. Combining these pathways is intended to produce weight loss beyond what single- or dual-receptor agonists achieve. Researchers attribute the observed potency to simultaneous engagement of all three targets, though the exact contribution of each receptor to overall effect remains under investigation.
Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.
Retatrutide is a synthetic peptide developed as a single molecule that activates three distinct hormone receptors: GLP-1, GIP, and glucagon. The compound carries the internal designation LY3437943 and was engineered by modifying the backbone of glucose-dependent insulinotropic polypeptide. Its sequence incorporates non-natural amino acids and a fatty acid side chain that extends circulation time. The triple-agonist design aims to combine appetite suppression, improved insulin response, and increased energy expenditure in one agent. Published reports describe it as an investigational product rather than an approved medicine.
| Property | Value | Notes |
|---|---|---|
| Receptor targets | GIP, GLP-1, glucagon | One molecule activates all three pathways |
| Research identifier | LY3437943 | Code used in published trial reports |
| Development stage | Phase 3, reported as ongoing | Not approved by any regulatory agency |
| Route studied | Subcutaneous | Administration form used in clinical trials |
| Studied populations | Adults with obesity or type 2 diabetes | Enrollment criteria vary between trials |
Acting as a triple agonist, the molecule binds the GLP-1, GIP, and glucagon receptors. GLP-1 activity slows gastric emptying and dampens appetite, while GIP signaling contributes to insulin sensitivity and fat metabolism. Glucagon receptor engagement raises energy expenditure and encourages fat breakdown, although it can also elevate blood glucose. Combining three pathways is intended to yield larger weight reduction than single or dual agonists, and researchers continue to examine how the balance among them shapes tolerability.
Clinical studies have reported notable reductions in body weight among participants. Early trials measured safety and explored several dose levels, and later studies tracked body-weight change over months of treatment. Investigators also monitor effects on glycemic markers, liver fat, and blood lipids. Because the compound is still in development, questions about long-term safety, cardiovascular outcomes, and durability after treatment ends remain open.
Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.
== Class C peptidomimetics == These structural mimetics include molecules that are highly modified when compared to their parent peptide sequence. Usually, a small-molecular scaffold is applied to project groups in analogy to the bioactive conformation of a peptide.
From March through July 2020, vitamin C was the subject of more US FDA warning letters than any other ingredient for claims for prevention and/or treatment of COVID-19. In April 2021, the US National Institutes of Health (NIH) COVID-19 Treatment Guidelines stated that "there are insufficient data to recommend either for or against the use of vitamin C for the prevention or treatment of COVID-19." In an update posted December 2022, the NIH position was unchanged:

Claude Monet (1840–1926), a resident of Le Havre from the age of five, in 1872 painted Impression soleil levant (Impression, Sunrise), a painting that gave its name to the impressionist movement. In 1867–1868, he painted many seascapes in the Le Havre region (Terrasse à Sainte-Adresse (Garden at Sainte-Adresse), 1867 Bateaux quittant le port (Boats Leaving the Port), 1874). The Musée Malraux houses some of his paintings : Waterlilies, London Parliament et Winter Sun at Lavacourt. Two other Impressionists, Camille Pissarro (1830–1903) and Maxime Maufra (1861–1918) also represented the port of Le Havre which also inspired Paul Signac (1863–1935), Albert Marquet (1875–1947), and Maurice de Vlaminck (1876–1958). Then came the school of Fauvism in which many artists did their training at Le Havre: Othon Friesz (1879–1949), Henri de Saint-Delis (1876–1958), Raoul Dufy (1877–1953), Georges Braque (1882–1963), Raymond Lecourt (1882–1946), Albert Copieux (1885–1956), who followed the course of the School of Fine Arts of Le Havre in the time of Charles Lhuillier. They left a number of paintings on the theme of the city and the port. In 1899, Henri de Toulouse-Lautrec (1864–1901) painted La serveuse anglaise du Star (The English waitress of Star) (Museum Toulouse-Lautrec, Albi) of a girl he met in a bar in the city. Other painters who painted Le Havre and/or its surroundings such as Sainte-Adresse can be cited in particular: Frédéric Bazille, John Gendall, Thomas Couture, Ambroise Louis Garneray, Pablo Picasso (Souvenir du Havre).
== Biosynthesis == Little gastrin I is synthesized from a 101-amino acid precursor protein called preprogastrin. A signal peptide of 21 amino acids is removed in the Endoplasmic reticulum, producing the 80-amino acid intermediate progastrin. Progastrin is then cleaved at specific site by Proprotein convertase as it moves through the Golgi apparatus toward secretory vesicles, producing gastrin-34 and gastrin-17 as the two major biologically active end products. In the normal human stomach, around 85% of amidated gastrin in the antral mucosa is in the G-17 form.
==== Extracellular proteases ==== The green microalgae Chlamydomonas coccoides and Dunaliella sp. and chlorella sphaerkii (a unicellular marine chlorophyte) were found to produce extracellular proteases. The diatom Chaetoceros didymus releases substantial amounts of proteases into the medium, this production is induced by the presence of the lytic bacterium Kordia algicida and is connected to the resistance of this alga against the effects of this bacterium. Some proteases are of functional importance in viral life cycles, thus being attractive targets for drug development.
=== Permethrin === Permethrin, a pyrethroid insecticide, is the most effective treatment for scabies, and remains the treatment of choice. It is applied from the neck down, usually before sleep, and left on for about 8 to 14 hours, then washed off in the morning. Care should be taken to coat the entire skin surface, not just symptomatic areas; any patch of skin left untreated can provide a "safe haven" for one or more mites to survive. One application is normally sufficient, as permethrin kills eggs, hatchlings, and adult mites, though many physicians recommend a second application three to seven days later as a precaution. Crusted scabies may require multiple applications or supplemental treatment with oral ivermectin (below). Permethrin may cause slight irritation of the skin that is usually tolerable. In recent years, concern has grown regarding the development of permethrin-resistant scabies. 2022 studies performed in Kuşadası, Turkey showed in cases where patients failed to improve with 5% permethrin, 100% of scabies mites analyzed were sensitive to in-vitro 5% permethrin, suggesting that lack of efficacy could be due to treatment non-compliance rather than true resistance. However, a 2024 study published in Salzburg, Austria revealed permethrin resistant-scabies in nearly 3 out of 4 cases studied. In this population, treatment with benzyl benzoate had a considerably higher cure rate. Additional studies will be needed to confirm the presence of permethrin-resistance and to determine its epidemiology.
Argon has approximately the same solubility in water as oxygen and is 2.5 times more soluble in water than nitrogen. Argon is colorless, odorless, nonflammable and nontoxic as a solid, liquid or gas. Argon is chemically inert under most conditions and forms no confirmed stable compounds at room temperature. Although argon is a noble gas, it can form some compounds under various extreme conditions. Argon fluorohydride (HArF), a compound of argon with fluorine and hydrogen that is stable below 17 K (−256.1 °C; −429.1 °F), has been demonstrated. Although the neutral ground-state chemical compounds of argon are presently limited to HArF, argon can form clathrates with water when atoms of argon are trapped in a lattice of water molecules. Ions, such as ArH+, and excited-state complexes, such as ArF, have been demonstrated. Theoretical calculation predicts several more argon compounds that should be stable but have not yet been synthesized.

AMPK probably also plays a role in increased peripheral insulin sensitivity, as metformin administration increases AMPK activity in skeletal muscle. AMPK is known to cause GLUT4 deployment to the plasma membrane, resulting in insulin-independent glucose uptake. Some metabolic actions of metformin do appear to occur by AMPK-independent mechanisms, however, AMPK likely has a modest overall effect and its activity is not likely to directly decrease gluconeogenesis in the liver. Metformin has indirect antiandrogenic effects in women with insulin resistance, such as those with PCOS, due to its beneficial effects on insulin sensitivity. It may reduce testosterone levels in such women by as much as 50%. A Cochrane review, though, found that metformin was only slightly effective for decreasing androgen levels in women with PCOS. Metformin also has significant effects on the gut microbiome, such as its effect on increasing agmatine production by gut bacteria, but the relative importance of this mechanism compared to other mechanisms is uncertain. Due to its effect on GLUT4 and AMPK, metformin has been described as an exercise mimetic.
== Natural occurrence == Nicotine is a secondary metabolite produced in a variety of plants in the family Solanaceae, most notably in tobacco Nicotiana tabacum, where it can be found at high concentrations of 0.5 to 7.5%. Nicotine is also present in other tobacco species, such as Nicotiana rustica (in amounts of 2–14%). Nicotine production is strongly induced in response to wounding as part of a jasmonate-dependent reaction. Specialist insects on tobacco, such as the tobacco hornworm (Manduca sexta), have a number of adaptations to the detoxification and even adaptive re-purposing of nicotine. Nicotine is also found at low concentrations in the nectar of tobacco plants, where it may promote outcrossing by affecting the behavior of hummingbird pollinators. Nicotine occurs in smaller amounts (varying from 2–7 μg/kg, or 20–70 millionths of a percent wet weight) in other Solanaceaeous plants, including some crop species such as potatoes, tomatoes, eggplant, and peppers, as well as non-crop species such as Duboisia hopwoodii. The amounts of nicotine in tomatoes lowers substantially as the fruit ripens. A 1999 report found "In some papers it is suggested that the contribution of dietary nicotine intake is significant when compared with exposure to ETS [environmental tobacco smoke] or by active smoking of small numbers of cigarettes. Others consider the dietary intake to be negligible unless inordinately large amounts of specific vegetables are consumed." The amount of nicotine eaten per day is roughly around 1.4 and 2.25 μg/day at the 95th percentile.
The amount of alcohol on the breath can be measured, without requiring drawing blood, by blowing into a breathalyzer, resulting in a breath alcohol content (BrAC). The BrAC specifically correlates with the concentration of alcohol in arterial blood, satisfying the equation BACarterial = BrAC × 2251 ± 46. Its correlation with the standard BAC found by drawing venous blood is less strong. Jurisdictions vary in the statutory conversion factor from BrAC to BAC, from 2000 to 2400. Many factors may affect the accuracy of a breathalyzer test, but they are the most common method for measuring alcohol concentrations in most jurisdictions.
It is clear that using oxygen to completely oxidize glucose provides an organism with far more energy than any oxygen-independent metabolic feature, and this is thought to be the reason why complex life appeared only after Earth's atmosphere accumulated large amounts of oxygen.
==== United Kingdom ==== Prescriptions of Oxycodone rose in Scotland by 430% between 2002 and 2008, prompting fears of usage problems that would mirror those of the United States. The first known death due to overdose in the UK occurred in 2002.

It is a synthetic peptide triagonist that engages GIP, GLP-1, and glucagon receptors. Investigational compounds in this class are studied for metabolic conditions rather than for a single organ system.
No regulatory agency has approved it for any indication. It remains an investigational product studied within controlled clinical trial programs.
A dual agonist engages two receptor targets, typically GIP and GLP-1. A triagonist adds glucagon receptor activity, which is intended to influence energy expenditure alongside appetite and glucose handling.
Retatrutide is an investigational peptide that activates the GLP-1, GIP, and glucagon receptors at the same time. It is being studied for obesity and type 2 diabetes and has not been approved for clinical use. The internal code LY3437943 refers to the same molecule.
