Retatrutide is a synthetic peptide developed as a single molecule that activates three distinct hormone receptors: GLP-1, GIP, and glucagon. The compound carries the internal designation LY3437943 and was engineered by modifying the backbone of glucose-dependent insulinotropic polypeptide. Its sequence incorporates non-natural amino acids and a fatty acid side chain that extends circulation time. The triple-agonist design aims to combine appetite suppression, improved insulin response, and increased energy expenditure in one agent. Published reports describe it as an investigational product rather than an approved medicine.
Each receptor contributes a different physiological effect. Activation of the GLP-1 receptor slows gastric emptying and reduces appetite signaling in the brain. GIP receptor activity influences insulin secretion and lipid handling, while glucagon receptor stimulation raises energy use and fat oxidation. Combining these pathways is intended to produce weight loss beyond what single- or dual-receptor agonists achieve. Researchers attribute the observed potency to simultaneous engagement of all three targets, though the exact contribution of each receptor to overall effect remains under investigation.
Randomized studies of retatrutide measure change in body weight as a percentage of baseline, along with absolute weight loss. Glycemic endpoints include hemoglobin A1c and fasting plasma glucose. Investigators also track blood pressure, lipid fractions, and liver fat content to characterize effects beyond weight alone. Trial designs typically use double-blind, placebo-controlled groups with periodic dose escalation, and they record adverse events throughout both treatment and follow-up periods.
Quantification of the peptide in biological samples generally relies on liquid chromatography coupled with tandem mass spectrometry. This approach separates the analyte from matrix components and detects it by mass-to-charge transitions specific to the molecule. Immunoassays offer higher throughput but can cross-react with related peptides and metabolites, so mass spectrometric methods are preferred when structural confirmation is required. Method validation typically addresses accuracy, precision, selectivity, and stability under handling conditions.

==== Structure ==== The myophosphorylase structure consists of 842 amino acids. The molecular weight of the unprocessed precursor is 97 kDa. The three-dimensional structure has been determined for this protein. The interactions of several amino acids in myophosphorylase's structure are known. Ser-14 is modified by phosphorylase kinase during activation of the enzyme. Lys-680 is involved in binding the pyridoxal phosphate, which is the active form of vitamin B6, a cofactor required by myophosphorylase. By similarity, other sites have been estimated: Tyr-76 binds AMP, Cys-109 and Cys-143 are involved in subunit association, and Tyr-156 may be involved in allosteric control.
In North America, the common name for the remains of a cigarette after smoking is a cigarette butt. In Britain, it is also called a dog-end or a fag end. The butt is typically about 30% of the cigarette's original length. It consists of a tissue tube which holds a filter and some remaining tobacco mixed with ash. They are the most numerically frequent litter in the world. Cigarette butts accumulate outside buildings, on parking lots, and streets where they can be transported through storm drains to streams, rivers, and beaches. In a 2013 trial, the city of Vancouver, British Columbia, partnered with TerraCycle to create a system to encourage the recycling of cigarette butts. A reward of 1¢ per collected butt was offered to determine the effectiveness of a deposit system similar to that of beverage containers.
== Origin of eukaryotes == The great oxygenation event began with the biologically induced appearance of oxygen in the Earth's atmosphere about 2.45 billion years ago. The rise of oxygen levels due to cyanobacterial photosynthesis in ancient microenvironments was probably highly toxic to the surrounding biota. Under these conditions, the selective pressure of oxidative stress is thought to have driven the evolutionary transformation of an archaeal lineage into the first eukaryotes. Oxidative stress might have acted in synergy with other environmental stresses (such as ultraviolet radiation and/or desiccation) to drive this selection. Selective pressure for efficient repair of oxidative DNA damages may have promoted the evolution of eukaryotic sex involving such features as cell-cell fusions, cytoskeleton-mediated chromosome movements and emergence of the nuclear membrane. Thus, the evolution of meiotic sex and eukaryogenesis may have been inseparable processes that evolved in large part to facilitate repair of oxidative DNA damages.

=== Catholic === American Order of United Catholics - Founded in January 1896 in New York City by Catholics who wished to counter the influence of the American Protective Association. It was "expected of the founders" that they would demand candidates for office who disapproved of the APA or other organizations that sought to discriminate against Catholics. A Supreme Council was organized on March 7, 1896, and the Order was organized "upon the usual secret society lines". They issued a circular that proclaimed that the Church did not oppose secret societies, except those that were oathbound. Catholic Aid Association - Founded in 1878 by German Catholics in Minnesota. The order began with 464 members from 10 parishes and was called the Deutsche Römisch-Katholische Unterstützungs-Gesellschaft von Minnesota; adopted its present name in 1923. Had 58,722 members in 1965, and approximately 78,000 in 1979. Headquarters in St. Paul, Minnesota. Local groups are called "subordinate Councils", of which there were 240 in 1979. Annual convention called the "Grand Council". Has rituals for initiation, installation of officers, and other purposes. Open to Roman Catholics 16-65 who are not a member of a secret society condemned by the Church. Sponsors matching grants program for Catholic elementary schools and religious education programs. Also, a College Tuition Scholarship Program has helped 800 CAA members receive degrees. Also sponsors banquets, family outings, dances, and youth activities; now Catholic United Financial.
The surgeon fabricates a metal foil template derived from the dimensions of the nasal wound. Applying a Doppler ultrasonic scanner, the surgeon identifies the axial pedicle of the tissue-flap (composed of the supraorbital artery and the supratrochlear artery), usually at the base, next to the medial brow; the point usually is between the midline and the supraorbital notch. Tracing the Doppler pulse of the blood flow of the supraorbital artery as far as possible, its delineation is continued as a vertical line, until it intersects with the hairline of the patient. The line extended from the pulse of the blood flow is the central axis of the forehead flap. The length of the flap is determined by placing an un-folded, un-stretched 4 × 4-inch gauze upon the wound, and with it measuring from the pedicle base to the distal (farthest) point of the wound. This measure is the length of the central axis of the skin flap. The template is rotated 180 degrees and placed over the distal (far) portion of the axis of the skin flap; the surgeon outlines it with a surgical marker. The outline markings are continued proximally and parallel to the central axis, maintaining a 2-cm width for the proximal flap. Without applying an injection of anaesthetic epinephrine, the flap is incised (cut), and the distal one-half is elevated between the frontalis muscle and the subcutaneous fat. At approximately the mid-portion of the forehead, the surgeon deepens the plane of the dissection down to the submuscular plane.
In June 1997, Zhang Hongchao was studying at Henan University of Economics and Law when he opened a shaved‑ice stall in Zhengzhou using a CN¥3,000 loan from his grandmother. The stall's early success convinced him to expand, and in 1999 he opened a second stand called Mixue Bingcheng. Due to a lack of experience, Hongchao had closed three stores in a little more than a year. Following the closure of his third store, he went to Hefei to sell tanghulu, which also failed. In spring 1999, Hongchao returned to Zhengzhou to start a new cold drink shop, though this would also be demolished; the failures of these street stalls were attributed to poor locations and ongoing urbanization.

Retatrutide is an investigational peptide that activates the GLP-1, GIP, and glucagon receptors at the same time. It is being studied for obesity and type 2 diabetes and has not been approved for clinical use. The internal code LY3437943 refers to the same molecule.
Most approved incretin therapies target one or two receptors, while retatrutide engages three. The addition of glucagon receptor activity is intended to raise energy expenditure. This broader mechanism is the subject of ongoing research rather than established practice.
No regulatory agency has approved retatrutide for any indication as of available public information. Development remains in later-stage trials. Availability claims found online should be treated as unverified.
Trials measure percentage change in body weight, absolute weight loss, and glycemic markers such as hemoglobin A1c. They also record blood pressure, lipids, and liver fat. Adverse events are tracked throughout.
